A Study on the Change of Enzyme Activities in Some Organs of Rat by Feeding of Diet Added Phenylmercuric Acetate or Methionine


Abstract
To observe the metabolic changes in organs of rat by feeding of diet with phenylmercuric acetate or methionine, the activities of alkaline phosphatase, glutamic pyruvic transaminase(G. P. T.) and glutamic oxaloacetic transaminase(G. O. T.) in the liver, kidney, small intestine and serum of rat were determined.
A total 170 male albino rats from pure strain, weighing 179-271g, were used for the experimental animals. They were divided into four groups as follows; 1. standard diet group. 2. fasting group contained 1-day, 2-day and 4-day fasting group. 3. feeding group of the high carbohydrate diet added methionine, phenylmercuric acetate, and methionine and phenylmercuric acetate, respectively. 4. feeding group of the high protein diet added methionine, phenylmercuric acetate, and methionine and phenylmercuric acetate, respectively. And the groups of 2 and 3 mentioned above were subdivided into three groups; 10-day, 20-day and 30-day feeding group, by feeding periods.
The results from this study are as follows:
1. Alkaline phosphatase activities in the liver and serum of fasting groups show a declining tendency according to fasting periods, but in kidney show a rising tendency and in small intestine, a declining tendency from 1-day fasting group to 2-day fasting group, on the contrary, rised to 67% at 4-day fasting group, compared with that of standard diet group.
2. G. P. T. activities in each organ of fasting groups has no great change from 1-day fasting to 2-day fasting, but rised to more than 100% at 4-day fasting group, compared with that of standard diet group. while G. 0. T. and G. P. T. activities are contrary to each other.
3. Alkaline phosphatase activities in liver is less in the feeding group of diet supplemented with methionine and mercury than in that supplemented mercury.
4. Alkaline phosphatase activities in kidney of high protein diet groups is less in addition of mercury and methionine than in addition of mercury only, but that of high carbohydrate diet groups is contrary to high protein diet groups.
5. G. P. T. activities in liver of high protein diet groups show a declining tendency according to adding each drug, but show a rising tendency in kidney, and in small intestine, a rising tendency from 1 day to 10 days, after that time decreased. This tendency made a marked difference in periods adding methionine, compared with that of control groups.
6. G. 0. T. activities in liver and kidney of each diet group rised generally by adding of each, compared with that of control group.
As a result of above, alkaline phosphatase activities in organs of high protein diet groups are less in addition of mercury and methionine than those in addition of mercury only, G. P. T. and G. 0. T. activities decreased by feeding of diets adding drugs, compared with that of control groups.
